What is NMN and how does it work?

NMN is a naturally occurring substance present in all life forms. NMN is important because it increases levels of NAD+, which is a key molecule in energy production. The process of ageing, lower energy, cell and tissue damage are all intrinsically linked to a decrease in NAD+. NAD+ is also needed to activate special proteins in the cells, called sirtuins, that keep our DNA healthy.

NAD+ itself is not easily absorbed into cells, but research has shown that when NMN is taken orally it is quickly absorbed and is the best way to boost NAD+.

What are the benefits of NMN?

By boosting cellular energy production, NMN can increase both physical and mental energy levels. It helps to improve cognitive function as well as sleep. In some people, it even helps lessen the effects of hangovers.

As a longevity and anti-ageing supplement, NMN can not only help keep your skin looking great, but beneath the surface it works to repair your DNA and help manage blood sugar levels.

    The need to know

    What's in the NMN supplement?

Each capsule contains 500mg of 99.5% pure B-nicotinamide mononucleotide (“NMN”). NMN is a derivative of a B vitamin. There are no fillers or bulking agents. The capsules are non-GMO and are suitable for vegans and vegetarians.

Our jars contain a month’s supply of 30 capsules. If you add NMN to your 4 weekly supplement pack you will receive 4 weeks’ supply (28 capsules).

    When to take it?

NMN can be taken before exercise or at any time during the day to boost resilience, drive and energy.

Athletes have reported improved sensation of energy when taken 20-30 minutes before exercise. This reflects studies that show NMN is absorbed with 5-10 minutes into the blood stream. NMN also passes the blood brain barrier to increase NAD+ levels in the brain, which improves cognitive function.

Taking it in the late afternoon has also been shown to improve sleep.

    What are the benefits?

NMN can bring the following benefits:

    • Increased energy. Research indicates that NMN is the best way to boost levels of NAD+, which is a key molecule in producing energy. As we age our body produces lower amounts of NAD+. This in turn affects a lot of systems in the body. A randomised control trial published in 2021 of 48 young and middle-aged trained runners were given NMN and trained 5-6 times a week with each session being 40-60 minutes. At 6 weeks their aerobic capacity was tested. It was shown that the aerobic capacity was increased in the NMN group against the placebo.
    • Improved cognitive function. A 2019 mice study showed that cerebral blood flow, which has an essential role in maintenance of healthy brain function, was improved by NMN. Blood flow is essential for a washout of harmful metabolites and to deliver oxygen and nutrients to the energy-hungry brain, given that the brain itself has limited energy reserves. The study also noted that NMN activated sirtuin genes and increased biogenesis of mitochondria (the body’s energy producers). Both of these help to reduce the cognitive decline of old age.
    • Longevity. By restoring NAD+ levels we can reduce, delay, or even reverse age-related diseases. The main benefits appear to be in chronic conditions such as diabetes, cognitive impairment, and cardiovascular conditions.
    • Better sleep. A Japanese study published in 2022 looked at 108 older adults and assessed their sleep when taking NMN.  Significant improvements were found in both the quality and duration of sleep. When taken in the afternoon it significantly reduced fatigue and improved lower limb function.
    • DNA repair. NMN activates DNA repair mechanisms (sirtuins) in cells. This means that cells behave like they should do when you are young. This is important because a gradual decline in DNA quality leads to cells becoming inert zombie cells with reduced function. When cells stop working properly problems occur: skin becomes wrinkled, brain connections are lost, and liver and kidney cells lose function.

    Can I get NMN naturally?

Although NMN is found in foods such as broccoli, cucumber and avocado, it is present in very small amounts (around 0.25-1.9 mg per 100g). In order to get the same amount of NMN as one 500mg capsule you would need to consume a kilo of broccoli every day.

The best way to boost NMN / NAD+ is to take a supplement.

    Is NMN safe?

A 2019 study studied the effects of NMN on 10 healthy Japanese men. They showed that daily doses of up to 500mg are safe.

Many leading longevity experts such as Prof. David Sinclair take up to 1g per day.

Multiple studies have been done on humans with doses up to 1200mg. None of these have noted any side effects.

NMN is recognised as an active product that is non-pharmaceutical as it exists in nature and is  generally recognised as safe (“GRAS”).

We recommend 1-2 capsules per day.

What other supplements work with NMN?

To boost the longevity effects of NMN and improve activation of the key anti-ageing pathways see our blogs on NMN and on Longevity.

We think the best Longevity supplements are as below. To make life easy we have put together in our Longevity pack.

    1. NMN The humanpeople NMN is 99.5% pure B-nicotinamide mononucleotide, which is highly absorbable in the gut. It increases energy production, increases AMPK and activates sirtuins.
    2. Quercetin increases NAD+ levels through inhibition of CD38, the enzyme that breaks down NAD+, thereby boosting the effects of NMN
    3. Resveratrol works by activating sirtuin pathways. Trans-resveratrol is the most active form and works synergistically with NMN.
    4. Coenzyme Q10 is a commonly used cofactor as it is as essential mitochondrial electron transporter as well as acting as an antioxidant. It delays brain atrophy and a blinded study showed that within 8 days the participants on CoQ10 were able to cycle further with lower fatigue. Synthesis of CoQ10 is dependent on vitamins B2 (riboflavin) and vitamins B6 (pyridoxal-phosphate), folate (vitamin B9), pantothenic acid (vitamin B5), niacin (vitamin B3) and vitamin C.
    5. D3 and K2
    6. B complex methylated active form

Whats the difference between niacin (vitamin B3) and NMN?

To increase NAD levels and activate the important sirtuin pathways it is beneficial to take NMN rather than niacin.

The reason for this is that nicotinamide NAM is a step further back in the process. It needs to be converted into NMN and then NAD+.

Taking NAM will inhibit the sirtuins. When sirtuin dependent deacetylase are activated by NAD+ the NAD+ is then converted into NAM. If there is too much NAM present it inhibits this step and you lose these benefits.

Vitamin B3, niacin and nicotinamide also cause flushing when taken at high doses.

Can NMN really help with weight management?

Studies in mice have shown that NMN increases metabolism, which includes the breakdown of fat. A different study, however, which looked at menopausal women did not replicate this finding, but it is worth noting that the dosages used in this study (250mg per day) were low.

We believe that a daily NMN dosage of 500mg can help with weight loss. A recent SIRT diet, which uses diet and supplementation to activate a number of sirtuin pathways, has been shown to lead to weight loss. It is claimed that the musician Adele used this to lose over 50 pounds.

To be clear, we are not marketing NMN as a weight loss supplement. If weight loss is the goal, NMN should be taken in conjunction with a healthy diet and lifestyle regime.

    Is NAD+ better than NMN?

They are different things. In short, NMN is the fastest way to boost NAD+. To boost NAD+ levels it needs to transported into the cells and then to the mitochondria.

To date no transporter has been found for NAD+. It appears that NAD+ has to be broken down into NMN or NR, transported into the cell and then converted back into NAD+.

In 2018 Grozio et al identified a transporter, Slc12a8, that moves NMN into the cell.

    The science behind NMN

    Our NMN is certified >99%

50% drop in NAD+ levels by the age of 45

From our mid-twenties the energy batteries in our cells (mitochondria) start to drop off in function due to a lack of NAD+. As we reach middle age this reduces to around 50%. To restore that youthful vitality, it is essential to support and boost the production of NAD+. Low levels of NAD+ are intrinsically linked to the signs of ageing: mental decline, fatigue, poor glucose control and skin quality.

NMN restores NAD+ in the cells

NMN increases levels of NAD+, which is a key molecule in energy production. The process of ageing, lower energy, cell and tissue damage are all intrinsically linked to a decrease in NAD+. NAD+ is also needed to activate special proteins in the cells, called sirtuins, that keep our DNA healthy.NAD+ itself is not easily absorbed into cells, but research has shown that when NMN is taken orally it is quickly absorbed and is the best way to boost NAD+.

    NMN and sirtuins

Professor David Sinclair of Harvard University is one of the world’s leading anti-aging researchers. His research on resveratrol, NAD, and sirtuins has been widely published and accepted as groundbreaking in the field of anti-ageing.

    What are sirtuins?

Sirtuins are a set of proteins with very important roles in the cell. In mammals there are 7 sirtuins labelled SIRT 1 to SIRT 7. To date SIRT1 and SIRT6 appear to the most important.

They are associated with the production of energy in the mitochondria and keeping DNA healthy. They also have a key role in suppressing cancerous tumours. Activating them is a good thing for increasing healthy lifespan. Critically, they consume NAD to function. By manipulating the expression of sirtuins it is possible to decrease and even reverse cell ageing.

Professor Sinclair has shown that supplementation with NMN and resveratrol activates the sirtuins, thereby reducing biological age.

    Third party tested

Our NMN has been third party tested in the UK by an independent ISO certified laboratory. It is >99% B-nicotinamide mononucleotide.

    The Science

    Clinically proven benefits of NMN

    Improved energy

To increase NAD levels and activate the essential sirtuin pathways it is beneficial to take NMN rather than niacin.

This is because nicotinamide NAM is a step further back in the process. It needs to be converted into NMN and then NAD+.

Taking NAM will inhibit the sirtuins. When sirtuin-dependent deacetylase are activated by NAD+ the NAD+ is then converted into NAM. If there is too much NAM present it inhibits this step and you lose these benefits.

Vitamin B3, niacin and nicotinamide also cause flushing when taken at high doses.

    Brain health

    Longevity

When taken in the afternoon, it significantly reduced fatigue and improved lower limb function.

    DNA and cell health

NMN activates DNA repair mechanisms (sirtuins) in cells. This means that cells behave like they should do when you are young.

This is important because a gradual decline in DNA quality leads to cells becoming inert zombie cells with reduced function. When cells stop working properly problems occur: skin becomes wrinkled, brain connections are lost, and liver and kidney cells lose function.
